The influence of culture system on the success of bovine embryo production in vitro | Vplyv systému kultivácie na úspešnosť produkcie bovinných embryí in vitro

The aim of our study was to compare the effect of two currently used culture systems on success of bovine embryo production after in vitro fertilization. Zygotes following in vitro maturation and fertilization were cultured in SOF medium without adjustment of O2 content in atmosphere (SOF), with 5% O2 (SOF-5) or in B2 medium in co-culture with BRL cell monolayer (B2-BRL). Embryo development was estimated on the basis of embryo cleavage rate (Day 2, D2) and blastocyst rate (Day 7, D7). The highest cleavage rate was obtained in groups SOF-5 (64.9%) and B2-BRL (63.6%) compared to SOF-20 (47.7%). The highest blastocyst rate was reached in B2-BRL group (28.3%), the lowest one was observed in SOF group (6.1%), whilst in the SOF-5 group a blastocyst yield was 14.4%. These results show that a more successful system of bovine embryo production in vitro can be obtained, even despite some disadvantages, by using the system of co-culture with BRL cell monolayer.
