Влияние оксида азота на внутриклеточную концентрацию ионов кальция в трансгенных растениях Nicotiana Plumbaginifolia

The interaction between NO metabolic system and [Ca 2+]-signal transduction with help of transgenic apoaequorin-expressed Nicotiana Plumbaginifolia plants in normal environment and in the oxidative stress conditions was analyzed. The 14-days germinants of Nicotiana Plumbaginifolia plants, expressing Ca2+ sensitive apoaequorin protein in the leaf cytoplasm were researched. Effect of nitric oxide donor sodium nitroprusside on cytosolic free calcium concentration ([Ca 2+]cyt) was revealed. The percentage of the integral increase of [Ca 2+]cyt was calculated and displayed. For the first time NO was shown to evoke dose-dependent cGMP-mediated [Ca 2+]cyt increase. Exogenous NO was found to reduce H2O2 influence on plants decreasing oxidative stress-induced Ca2+ response. The inhibitor analysis showed that endogenous NO responsible for the reduction of H2O2-evoked Ca2+ response occurs with the involvement of NOS-like plant enzyme sensitive to the oxidative stress. It was revealed that NO protect the plants against the oxidative stress, declining H2O2-induced Ca2+ answer.
