cDNA cloning of bZIP protein HY5 in tsuda tumip and its transcript expression analysis

صينى. HY5是一类碱性亮氨酸拉链类型的转录因子。在拟南芥中它是光形态建成和光诱导基因表达的正调控因子。为了研究津田芜菁HY5在花青素依光性合成过程中的表达特性,我们通过同源克隆得到了编码津田芜菁HY5的cDNA序列,长504bp,编码167个氨基酸,含有bZIP功能域结构,与拟南芥中的HY5同源性达88%,命名为BrHY5。Northern杂交分析表明BrHY5的表达没有组织特异性,在整株幼苗的表达不受光特异诱导,而在开花期的肉质根表皮中受UV―A诱导表达,但随处理时间延长差异不显著。这表明BrHY5可能作为转录因子,在津田芜菁幼苗中组成型表达,为光形态建成所必需;在形态建成结束的开花期时的大多器官组织几乎不表达,而在肉质根表皮中受UV―A诱导表达。
اظهر المزيد [+] اقل [-]إنجليزي. HY5, a bZIP transcription factor, is photomorphogenesis and positive regulator of light-induced gene expression in Arabidopsis. To study the expression character of HY5 in pigment light-dependent Brassica Rapa "Tsuda", the homolog cDNA with 504 base pairs of HY5 in "Tsuda" was isolated and designated BrHY5. BrHY5 contains open reading frames that encode deduced protein with 167 amino acid residues including bZIP domain and shows 88% sequence identity to Arabidopsis HY5. Northern hybridization experiments revealed that the expression of BrHY5 had no tissue specificity and had almost no difference in seedlings under light treatment, but its expression is specificity in epidermis of the swollen hypoeotyls under UV-A light. These observations indicate that the expression of bZIP transcription factor BrHY5 is constitutive in seedlings for photomorphogenesis and is almost not detected in most tissue and organ of the period of anthesis but in epidermis it is induced by UV-A.
