صينى. 为建立醋酸甲羟孕酮(MPA)快速准确的免疫学分析技术,对MPA人工抗原的合成鉴定进行研究。将MPA肟化后与琥珀酸酐反应制得MPA半琥珀酸(MPA-HS),然后通过碳二亚胺法将MPA-HS与牛血清白蛋白(BSA)偶联合成人工抗原MPA-BSA,并采用紫外全波长扫描法、SDS-PAGE法、ELISA检测法和动物免疫试验对其进行鉴定。MPA与BSA的偶联比为8∶1时,合成的人工抗原MPA-BSA具有良好免疫原性与反应原性。成功合成了人工抗原MPA-BSA,为进一步的MPA免疫学分析技术研究奠定了基础。
اظهر المزيد [+] اقل [-]إنجليزي. This study was to develop a rapid and sensitive technology for the detection of Medroxyprogeste rone Acetate(MPA). The atrificial antigen of MPA was synthesized and identified in the study. MPA hemi hemi-succinate (MPA-HS) was produced by the oximation reaction of MPA with the reaction between oximinooximated-MPA and with succinic anhydride, then the hapten MPA-HS was conjugated to protein carrier BSA by using the coupling agent diisopropylcarbodiimide, and the synthesis synthesized antigen MPA-BSA was identified by full-wavelength the ultraviolet full wavelength scanner, SDS-PAGE, ELISA test and animal-inoculation immunization test. The results showed that the synthesis synthesized antigen MPA-BSA with good immunogenicity and reactogenicityantigenicity, and the optimized conjugation molecular ratio of coupling reaction was determined to be 8∶1.
