صينى. [目的]研究西兰苔籽中异硫氰酸盐的抗肿瘤活性。[方法]利用GC-MS联用仪对西兰苔籽中的异硫氰酸盐进行了分析和鉴定,并通过MTT试验和相差显微镜观察研究了其抗肿瘤活性。[结果]通过GC-MS分析从西兰苔籽提取物中成功鉴定出5种异硫氰酸盐,占挥发性物质的80%。体外细胞毒性试验表明,异硫氰酸盐能有效抑制卵巢癌细胞OVCAR-3的生长,且抑制率随药物浓度的增大而增大,呈明显的剂量关系。异硫氰酸盐浓度为78.3 μg/ml时,对OVCAR-3细胞生长的抑制率为50%。对照组的OVCAR-3细胞生长良好,低浓度加药组(10、30、60 μg/ml)OVCAR-3细胞的形态和数目发生了变化,圆形脱壁细胞逐渐增多。随着药物浓度的继续增加,OVCAR-3细胞的形态变得很不规则,凋亡小体出现。[结论]该试验为我国天然抗癌药物研究奠定了基础。
اظهر المزيد [+] اقل [-]إنجليزي. [Objective] The aim was to research the antitumor activities of isothiocyanates from broccolini seeds. [Method] The isothiocyanates from broccolini seeds were analyzed and identified with GC-MS instrument and their antitumor activities were researched through MTT experiment and observation with phase contrast microscope. [Result] Five kinds of isothiocyanates were identified successfully from the extracts from broccolini seeds by GC-MS analysis and they accounted for 80% in volatile substances. It was shown in cytotoxicity test that the growth of ovarian cancer cell OVCAR-3 could be inhibited by isothiocyanates effectively and its inhibitory rate was increased along with the increase of its concn.,showing obvious dose relationship. When the concn. of isothiocyanates was 78.3 ug/ml,its inhibitory rate on the growth of OVCAR-3 cells was 50%. The OVCAR-3 cells in the control group grew well. In the groups added with isothiocyanates at low concn. (10, 30 and 60 ug/ml),the shape and quantity of OVCAR-3 cells got changed and the round digested cells were increased gradually. As the concn. of isothiocyanates was increased continuously,the shape of OVCAR-3 cells was changed to be quite irregular and the apoptotic bodies appeared. [Conclusion] This experiment laid a foundation for the research on natural anticancer drug in China.
