Sperm cryopreservation involves major changes that will affect negatively the sperm quality obtained after thawing. These changes are mainly due to thermal and osmotic stress. Susceptibility of sperm to chilling injury has been related to the sperm membrane composition. The aim of this study was to determine whether individuals with greater resistance to freezing and thawing process may present also greater resistance to cold shock as a first step to develop a test to select the ejaculates worth of being cryopreserved. The cluster analysis classification according to the values of percentage of total motile sperm after thawing split ejaculates of different males into two groups ("good" and "bad freezers"). When these groups were used to evaluate the sperm quality after ejaculates were subjected to cold shock, we observed that sperm from males classified as "good freezers" exhibited also higher percentages of total motile sperm (74.96+/-4.49%) than the sperm from "bad freezers" (55.57+/-5.80%). According to our results, the sperm response to cold shock (in terms of total motile sperm) could have a relationship with the percentage of total motile sperm obtained after thawing, although the sperm membrane integrity does not seem to be involved. Nevertheless, further studies are needed to assess the usefulness of this cold shock resistance test as a possible predictor of sperm quality after cryopreservation.
