The aim of this study was to assess the effects of different cryoprotectans on quality parameters of frozen-thawed rabbit semen. In the first experiment, six extenders trisbased were compared: A (3.5M DMSO+0.1M sucrose), B (1.5M acetamide), C (3M DMSO+0.1M sucrose +1.5M acetamide), D (3M DMSO+0.1M sucrose+1M acetamide, E (2.5M DMSO+0.1M sucrose+1.5M acetamide, F (2.5M DMSO+0.1M sucrose+1M acetamide). In a second experiment two extenders were compared: D and G (3M DMSO+0.1M sucrose+10% dextran). Motility was assessed using CASA and acrosomal integrity and viability were evaluated using flow cytometry. No significant differences were found between the six freezing extenders when motility was considered, while viability and acrosomal integrity were lower in the B medium than in the other extenders. The presence of dextran improved the percentage of live-acrosome intact sperm. However, it is necessary to evaluate the membrane stabilization effect of dextran on in vivo fertility and prolificacy.
