In vitro systems that simulate rumen fermentation should allow for a microbial growth similar to what occurs in the rumen. Althrough these systems have been widely used over the last 20 years, there is little information about the evolution of the microbial communities compared with the rumen. Furthermore, these systems have the restriction of high pressure that accumulates in the jars. The aim of this work was to study the bacterial growth in the Ankom sup (RF) in vitro fermentation system over 24 hours using rumen inoculum from goats and a diet based on alfalfa hay and oats (1:1). Total bacteria were quantified by using qPCR on DNA extracts of jars' content at 4 hours intervals along the 24 hours of incubation and of the inoculum. Results showed that the use of the rumen liquid fraction as inoculum involves a decrease in the original bacterial biomass. However, the decrease is fully compensated by a rapid bacterial growth during incubation, reaching a biomass equivalent to that in the original rumen liquid fraction. Nevertheless, the average concentration over the 24 hours of incubation was lower than in the rumen content, which might be considered for extrapolating in vitro results to in vivo conditions.
