Identification of Snps in the EXON 4 and 5 of POU1F1 gene and milk production traits in Sarda breed sheep

The genetic improvement of farm animals increasingly uses modern molecular biology techniques, in order to increase the productive traits (Dekker, 2004). Among the several genes involved in the regulation of the production processes, the POU1F1 gene (or also called Pit1 or GHF1) surely plays an important role. The aim of the present study was to detect the presence of mutation in the exons 4 and 5 of the POU1F1 nucleotide sequence and whether these substitutions could be related to milk yield and milk quality of dairy Sarda breed sheep. The research has been conducted on 140 ewes of Sarda breed from two farms with similar management conditions located in central Sardinia (39 deg 36 min N). Individual milk yield was recorded monthly and of each subject a milk sample was collected and analyzed to test the content of fat, protein, casein, lactose and somatic cell count (CombiFoss 6000, Foss Electric, Denmark). Genomic DNA was extracted from whole blood using a commercial DNA extraction kit (Puregene Blood Core Kit B, Qiagen Sciences, Marylad, USA). In conclusion it can be confirmed that the analyzed POU1F1 fragments do not influence the productive levels and milk traits in Sarda breed sheep. However, expanding the investigation on other parts of this gene, such as the promoter region, may provide additional information on other production traits in ovine species.
