Polymorphism of SLC11A1 gene in the Sarda goat

This research was carried out with the aim of analyzing SLC11A1 gene polymorphism in the Sarda goat and evaluates possible relationships with the composition of milk. SLC11A1 microsatellite was examined in 474 multiparous goats form 23 herds located in Sardinia. Milk yield was measured in the middle of lactation and a milk sample was taken to determine the percentage of fat lactose and proteins, pH, freezing point, SCC and TMC. Microsatellite genotyping was obtained by PCR-SSCP and DNA sequencing. Based on guanine-thymine repeats (GT)n in the 3'-UTR region (15th exon) of goat SLC11A1 gene, 8 alleles were detected, GT11, GT12, GT14, GT15, GT16, GT17, GT18 and GT19, combined in 24 different genotypes. The most frequent genotype was the GT16-GT16 (43.25%), followed by GT16-GT17 (12.87%), GT16-GT19 (10.76%) and GT14-GT16 (10.34%). Statistical analysis showed several relationships between genotype and milk yield and composition. In particular, the GT17-GT17 genotype showed the lowest milk yield and the highest protein content. No effect on genotype was noted, however, for the fat content, TMC and SCC. In conclusion this gene confirmed the great genetic variability of the Sarda goat, while the relationship with milk chemical, physical and cytological traits should be further investigated.
