Genotyping of RS-12 (S-12) mumps virus strain.

RS-12 mumps virus strain was isolated at the Razi institute in 1986. This strain isolated from throatwashing of a patient and developed to vaccine by serial passage of the pathogen. Although RS-12 isolated in over 20 years ago, only its very small sequences was previously determined. In order to investigate the genotype of the virus, sequence of SH gene of the low-passage virus was determined. The sequence of F and HN genes also were determined. For prediction and assessment of cross-reacting between this strain and others, the deduced amino acid sequence of HN and F proteins that comprise immunogenic epitopes were compared to other strains. Results clearly revealed that RS-12 belongs to genotype H. Phylogenetic analysis of entire SH gene showed that RS-12 is closely related (98. homology) to European (especially Russia, Belarus and UK) strains, and this genotype (H) has been spreading in this region of the world (middle east to Europe) for many years. The multiple sequence alignment revealed that neutralizing antibodies directed against HN glycoprotein of genotype H members, such as RS-12, could be potentially effective against most other genotypes. These data confirmed the previous results that RS-12 could be an effective vaccine in most parts of the worlds, especially in the Middle East and Central Asia countries. Further research on elucidating the molecular basis of attenuation of the RS-12 is essential.
