Classification of collected forage crops with molecular markers

In this investigation, genetic diversity of 20 barley (Hordeum sp) genotypes collected from various regions of country, using amplified fragment length polymorphism (AFLP) was studied. Seven AFLP primer combinations generated 195 polymorphic loci and showed extreme variability and genetic complexity among studied genotypes. The number of polymorphic loci varied from 17 to 37 for each primer combination. Polymorphic information content for each primer combinations ranged from 0.002 to 0.719. Cluster analysis using 195 polymorphic loci, separated the genotypes to seven distinct groups. Also, molecular analyses showed an extraordinary variation among the genotypes so that, the highest genetic similarity between Hb-1 and Hm-7 was only 0.55. Investigating considerable differences between the AFLP patterns of ecotypes despite a few number of utilized primer combinations proved a high level of differentiation between the genotypes. Hence, molecular study acceptably corresponded the morphological variation of ecotypes. Thus, this is evidently clear that the collected ecotypes are extremely valuable for further breeding programs in order to gain the stronger tolerant cultivars.
