The objective of this doctoral thesis was the identification of a marker linked to the Plasmopara-resistance gene Pl2 and the evaluation of different marker systems (isozymes, RFLPs, PCR based polymorphisms) for the characterization of genotypes. Two pairs of near isogenic sunflower lines which differ with respect to the Pl2 gene, F1-hybrids and the line HA89 (cms and fertile analogue) were analyzed. The following results were obtained: The calculation of genetic similarity of lines and hybrids was possible in principle by using isozymes, oligonucleotide-probes for DNA-fingerprinting and primers for PCR-analysis. The best system for the characterization of genotypes was DNA-fingerprinting with oligonucleotide-probes. The probe (GATA)4 showed bands which were qualitatively and quantitatively better than those obtained with probes (GACA)4 and (GGAT)4. Both (GATA)4 and (GGAT)4 hybridization patterns were specific for all near isogenic lines. The qualification of an enzyme/probe combination for generating specific fingerprints depended mainly on the probe selected and not on the enzyme. It was possible to identify the NlLs, the F1-hybrids and HA89 (cms and fertile analogue showed identical patterns) and to demonstrate the homogeneity of different sunflower lines by using the combination Dral/(GATA)4. Isozymes and PCR-primers were both suitable for the characterization of genotypes because their pattern showed the same degree of variability in the present case. The combination of two patterns allowed the identification of one pair of near isogenic lines and both F1-hybrids only. The non-radioactive Digoxigenin-system was an attractive alternative to the radioactive detection with alpha(32P)dCTP as long as there was no need to estimate the amount of detected DNA.
