Evaluation of proteinuria in diabetic patients by one dimensional sodium dodecyl sulfate- polyacrylamide gel electrophoresis.

The study of proteins which are most important constituents of human body has been always interesting to the scientists and investigators. The investigations on proteins have been primarily done on blood, but with regard to the fact the kidneys are involved in clearance of the plasma, therefore those components in some way are released into the urine. In spite of trace amount of proteins present in the urine, various techniques have been used to isolate and separate these proteins. The proteins isolated by these techniques have diferred from each other. The technique which has been used in the present dissertation is an electrophoresis technique and technique is very sensitive and can measure proteins precisely. Two kinds of electrophoresis technique used in this study were as follow : 1- Modified Laemmlie technique with tubular cast model. 2- Gradient gel concentration with O'farrell cast slab methods. According to our finding using tubular cast with gradient gel concentration has had other results. The studies conducted in our laboratory, the amount of uromucoid is less than the amount of albumin. Therefore in our studies the albumin have been regarded as the main protein which can be excreted in the urine. According to our finding diabetic patients have been divided into three groups: 1- In the first group the amount of albumin excreting was in the range of normal, in such patients, they were young and the course of the illness was short. 2- Second group the amount of albumin excretion was less than normal, but due to L. M. W. proteins excretion, is increased it is likely that due to the proteinuria in these patients, were results from tubular proteinuria. Third group the amount of albumin excreted in these patient was more than normal in this group of patients due to prolonged illness and an increase in age.
