The effect of different nutrients on enzyme production was studied with Amylomyces as strain. The media with a mixed mineral solution containing 0.05% of (NH4)2SO4, KH2PO4 and 0.17% MgSO4 was the best media for enzyme production. With this media enzyme activity was 31.41% higher than media with each of the following added: corn step liquor 3.5%, malt extract 1%, soybean meal 5%, K2HPO4 0.5%, KH2PO4 0.5%, MgSO4 0.5% and CaCl2 1%. The substitution of rice bran for wheat bran at the ratio of 25:75 or 50:50 in the media resulted in approximately the same amount of enzyme production as with wheat bran alone. With the wheat bran and water (1:1.2) media Amylomyces as produced high concentration of enzyme by the tray method at room temperature, as much as that by Amylomyces Ac, but Amylomyces as grew better and faster. The optimum inoculum was 3-5% and the difference in enzyme activity at day 4 and 5 was small, only 1%. The preservation of the produced enzyme solution stored at 4 Deg Cent. was a better method compared to storing enzyme solution with sodium benzoate at 0 Deg Cent. and 4 Deg. Cent. When stored as mold bran and enzyme solution at 4 Deg. Cent. for one month enzyme activity decreased 4.13 and 3,57% respectively.
