Enzyme polymorphism between Oryza sativa and O. minuta and its use to monitor introgression in O. sativa/O. minuta backcross derivatives.

Isozyme analysis of Oryza sativa, IR31917-45-3-2 and Oryza minuta accessions, Acc. 101089 and Acc. 101141, revealed interspecific variation in nine out of twelve enzyme systems. Information was obtained on the enzymatic and genetic properties of the O. minuta isozymes, such as quarternary enzyme structure and dosage, their possible associations and allelic relationships with known O. sativa alleles. Combined with chromosome data, the interspecific variation was exploited to monitor the relative genetic contribution of the two parents in the IR31917-45-3-2 / Acc. 101141 F1 hybrids and backcross progenies. The isozyme content of the F1 reflected its triploid nature (ABC genome composition) while that of the backcross progenies paralleled the loss of alien chromosomes during the backcrossing process. Presumptive recombinants or translocation lines (2n=24) and addition lines (2n=25) having one or two alien isozymes were identified as potential materials for correlation of alien isozymes with other phenotypes, e.g. morphological, physiological, and pest resistance properties. Progenies were also selected which are suitable for use in the production of near-isogenic lines of the loci encoding ENP-M1, EST-M2, MDH-M1, MDH-M2, MDH-M4 and MDH-M7. Those near isogenic lines would be of value for determining the chromosome location of Enp-1, Est-A, and Mdh Loci in O. sativa.
