Effects of auxin and cytokinin on formation of callus and shoot of in vitro culturing leaf explant in African violets [Saintpaulia ionantha].

The effects of 1-naphthaleneacetic acid (NAA) and 6-benzyladenine (BA) as plant growth regulators on callus formation, growth and shoot formation of leaf explants cultured in the African violet (Saintpaulia ionantha cv. Susie) were studied in vitro. Experimental plots consisted of 15 levels modified with Murashige and skoog (MS) basal medium supplemented with 0, 0.5, or 1.0 mg.liter(-1) NAA and 0, 0.05, 0.1, 0.5 or 1.0 mg.liter(-1) BA in combination. The leaf tissue explants were cultured 6 weeks on an incubator maintained at 25 +- 2 degrees C under cool-white fluorescent lamp (3.0 Klux) and a 16-hr photoperiod. The number of days from explants placed into solid medium to the beginning formation of callus, shoot and adventitious root, from callus formation to adventitious root were shortest in cultures containing NAA 0.5 mg and BA 0.5-1.0 mg.liter(-1) in combination. The number of shoot formation is maximized at cultured in combination NAA 1.0 mg: BA 1.0 mg.liter(-1). The adventitious root formation increased at cultures containing NAA 0.5 mg: BA 0.1 mg.liter(-1) in combination, while the higher concentration was inhibited the these. Cultures with NAA 0 mg: BA 0-1.0 mg.liter(-1) in combination were not formed the adventitious root. The total living organisms of callus and shoot were weightest at culturing with NAA 0.5 mg: BA 1.0 mg.liter(-1) in combination. The morphogenesis on in vitro culture in African violet leaf tissue explant showed good results on 0.5 mg.liter(-1) NAA and BA in combination, while these morphogenesis was effectiveness the composite than that single treatment of NAA and BA.
