Optimum amounts of bagged substrate for cultivating Pleurotus mushrooms were tried by applying sawdust, cassava root and water, 7: 3: 9. Substrate of 500 grams per bag in P. ostreatus which was significantly different from 100 grams per bag. In P. sajor-caju and P. cystidiosus, amounts of bagged substrate used had no effect on mushroom yields. Effect of water content in substrate on mushroom yields was also studied by adding three different types of water into it. Combinations used were sawdust, cassava root and water. There was no significant difference in yields of both P. ostreatus and P. sajor-caju. In P. cystidiosus, combination with six parts of water gave significantly highest yield. Incubation periods suitable for producing mycelial blocks were determined by allowing mycelia to grow on mixture of sawdust, cassava root and water. Four incubation regimes were applied as follows: at time when the mycelium permeated a half of bagged substrate, completely permeated whole substrate, completely permeated whole substrate plus 10 days and 15 days of further incubation. In P. ostreatus and P. sajor-caju, mushroom yields were not affected by incubation periods of mycelial block. In P. cystidiosus, periods at which mycelium permeated a half of bagged substrate and completely permeated whole substrate plus 10 days of further incubation periods of mycelial block. In P. cystidiosus, periods at which mycelium permeated a half of bagged substrate and completely permeated whole substrate plus 10 days of further incubation gave comparable higher yields.
