In vitro seed germination and seedling characterization of different transgenic events of papaya (Carica papaya L.)

More than 7000 seeds of transgenic Carica papaya that were produced and stored between the years 2003 and 2009 were recently evaluated for germination rate, seedling emergence and growth under in vitro conditions. Before the evaluation were performed, seed sterilization and pretreatment protocols were optimized to be able to germinate in vitro the seeds kept for 2 to 8 years. Because the seeds could not grow readily by standard sowing practices, in vitro germination using naked embryos was done to maximize the percentage of variable, undamaged seed. The objectives of this study were to measure the variability among stored seeds representing various transformation events across different generations and to determine how the various in vitro growth parameters relate to seedling establishment under greenhouse conditions. Emergence percentage, days for onset of germination, shoot size, and root length were among the variables measured. Germination rates were also correlated with these measures of seedling vigor. The range of response and other possible causes of variable performance among and within transgenic lines will be discussed.
