Papaya ringspot virus (PRSV) coat protein gene expression in advanced generation of selected PRSV-resistant papaya line in the Philippines

Selected T4 lines from candidate event 132 of transgenic papaya carrying the coat protein (CP) gene of PRSV that showed resistance to PRSV after mechanical inoculation were subjected to molecular analysis. The number of inserts was determined by Southern analysis. Preliminary Southern blots using a probe corresponding to a fragment of the CP gene show only one insertion point in the papaya genome. Expression of the PRSV CP gene in these selected transgenic lines was analyzed at the RNA and protein levels. The PRSV CP transcript was detected in the transgenic plants by RT-PCR using CP-specific primers. Previously reported data showed the presence of endogenous PRSV CP in transgenic plants. This was further confirmed by the presence of the CP product in the resistant lines as detected by western blot method using PRSV antiserum.
