Molecular and biological characterization of cucurbit aphid borne yellow virus (CABYV) causing the namamarako (syndrome) in ampalaya

A total of fourteen NMK virus isolates were obtained from different ampalaya growing areas of Laguna and the Central Luzon Region including Bulacan, Pampanga, Tarlac and Pangasinan. Each of the isolates were isolated and transmitted to healthy ampalaya seedlings by grafting and aphid transmission using Aphis gossyppii in persistent manner. Almost all of the isolates exhibited the typical NMK symptoms which only varied in disease severity. Virus infection in the area was estimated and observed to be higher in Bulacan with 75-90% infection while a lower rate of infection was seen in Laguna with 30-60 percent. Aphids have been used as an efficient vector for NMK transmission along with grafting. Four of the NMK virus isolates namely; Bulacan (2), Laguna (1), and Pangasinan (1) were propagated and maintained in ampalaya by aphids and graft inoculation inside insect proof cages. Two of the isolates, Laguna and Bulacan were used to inoculate some host plant species including plants from the family Chenopodiaceae, Aizoceae, Cucurbitaceae, Caricaceae and Solanaceae. Results of this study showed only 2 species of cucurbits (Zucchine and Luffa) to be susceptible to both NMK isolates. The total RNA from both NMK infected isolates were isolated and amplified by RT-PCR amplification yielding the expected 600 bp DNA fragment using a set of primers, CE9 and CE10 based on the published sequence of CATYV luteovirus and different PCR conditions.
