Comparative study of micropropagation techniques on Typhonium Flagelliforme

A micropropagation technique by means of aeration was tested on Typhonium flage/liforme and compared with other conventional micropropagation techniques such as the solid culture technique and shake culture technique. Aeration technique stimulates multiple bud formation more rapidly than solid culture technique and the shake culture technique. Shoots of T. flage/liforme produced 2.5 buds per shoot when cultured by aeration technique in 3 weeks compared with 1.6 buds per shoot by solid culture technique and only 1.2 buds per shoot by shake culture technique. Shoots of T. flage/liforme produced 4.0 buds, which is significantly more than multiple buds formed from tuber tissues. Shoots of T. flage/liforme produced the most multiple buds when cultured in modified liquid MS medium (X) with 40g/1 sucrose after 4 weeks in aeration tubes. In this liquid medium, light intensity of 800 lux is adequate for the highest multiple bud formation. This study als o showed that 5 explants in one aeration, tube are most suitable for multiple bud formation. The aeration rate of 25.3 ml/min is sufficient to produce high number of multiple buds.
