Characterization of serum proteins among Filipinos with normal to high cholesterol levels using SDS- polyacrylamide gel electrophoresis

SDS-PAGE of serum from 120 Filipinos with various cholesterol levels was conducted to assess genetic diversity and to identify possible genetic marker. Twenty-five band positions were observed with characteristic relative mobilities. The estimated MW (molecular weight) of the human sera protein bands ranged from or equal to 20 kDa to 220 kDa. The number of bands per sample ranged from 7 to 14, with 10 bands being the most frequent. The variation observed in the number of bands can be attributed to the differences of the individual samples. Some bands occur at very low frequencies (10%) in intermediate and normal groups. Round 6 is notably absent in samples from the high cholesterol group. Thus, there is no abnormal band observed among the high cholesterol group but rather absence of protein band 6 was noted. The protein profiles show only two protein banding profiles shared by a few samples, others are all unique and distinct per sample. Thus, the protein profile of Filipinos from the three groups of cholesterol levels cannot be established due to the heterogeneity of the groups analyzed. Kruskal-Wallis analysis of variance show no significant differences, indicating that the various groups cannot be distinguished based on the observed band frequencies. Cluster analysis shows several grouping based on the cholesterol groups, but none based on age or gender. There were several samples which have identical band pattern profiles as indicated by a similarity index of 1.0 in the dendrogram. However, the plot of probabilities of assignment of each sample to the three cholesterol groups show that the samples cannot all be assigned in the groupings identified based on their protein band profiles.
