The conditions for the comfrey root aqueous extract obtained by the Soxhlet extraction was defined in this paper. Ponomarev’s model was applied for modeling the extraction kinetics. The allantoin content in the extract was determined by HPLC method, and the total flavonoids content was measured by the spectrophotometric method using AlCl3 . The antioxidant activity of the extracts towards stable 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical was determined spectrophotometrically. Optimal conditions for obtaining the maximum yield of the total extractive matter by using the Soxhlet extraction (6.30 g/100 g of dry plant material) with a high content of allantoin were: hydromodule 1:10 m/v and the extraction time 240 minutes. The Allantoin content in an aqueous comfrey extract was 8.91 g/100 g of the dry extract. The content of total flavonoids in an aqueous comfrey root extract was 18.90 mg RE / g dry extract. The extent of DPPH radical scavenging, incubated by the extract in concentration of 0.15 mg/cm3 was over 80%, and slightly increases with the further increase of the extract concentration. With non-incubated extracts the extent of DPPH radical scavenging increases with the increase of the extract concentration. The non-incubated extract in the concentration of 0.4 mg/cm3 shows 77.3% of DPPH radical neutralization.
