Cloning, expression of alpha-amylase gene from oyster (Saccostrea commercialis)

Full length cDNA sequence of the alpha-amylase (ScAmy) gene from the oyster, S. commercialis, was identified using Rapid Amplification of cDNA End (RACE) technique. The gene composed of 1,759 bp containing an 1,563 bp open reading frame (ORF). The ORF encoded a deduced protein with 520 amino acid residues. The predicted molecular weight and isoelectric point (pI) of the mature protein were 59.9 kDa and 4.98, respectively. The ScAmy expression in specific tissues was studied by real-timePCR technique. The results showed that ScAmy was expressed in digestive gland with the highest levels and in stomach, palps, and mantle with moderate level. The lowest expression was found in gill and muscle.
