Mz-350 a broad-spectrum antimicrobial agent of streptomyces oriigen:taxonomy, fermentation, isolation,biological properties and characterization

A soil isolate of actinomycete strain MZ-350, was found to produce a promising antimicrobial agent. The presence of LL- diaminopimilic acid, glycine and non characteristic sugar pattern in the cell wall assigned this strain to genus Streptomyces. From the morphological, cultural and physiological characteristics, this strain was determined to be Streptomyces Iydicas. Thus, it was given the suggested name Streptomyces Iydicas, MZ-350. The active metabolite was extracted from the fermentation beer using ethyl acetate saturated with water (1:1 v/v) at pH 7.5. The separation of the active compound and its purification was performed using thin layer chromatography (TLC) and column chromatography (CC) techniques. The physicochemical properties of the purified compound viz. Color, melting point, solubility, elemental analysis, spectroscopic characteristics and chemical reactions have been investigated. This analysis indicates a suggested empirical formula of C. 2,H . 6N. O. .The antibiotic possesses a promising activities against all tested Gram positive bacteria and yeast but moderate (mailed) activities against both Gram negative bacteria and filamentous fungi. The moderate toxicity also was investigated (LD50= 55mg/ Kg). In conclusion, the collected data emphasized the fact that the purified active compound was suggestive of being belonging to piperdine alkaloid antibiotics produced by Streptomyce.Iydicas, MZ-350.
