Effect of isolation time and the sonicator's amplitude on the isolation of GSH from yeast by ultrasonic method

The effect of isolation time and amplitude of the sonicator on the isolation of GSH from baker yeast cells were studied. The yeast cells were disrupted by Ultrasonic Method. The disrupted cells solution was then centrifuged at 12,000 rpm for 20 minutes and the GSH content was analyzed by Spectrophotometer at 412 nm. The results obtained showed that the optimum value of the isolation time and the amplitude of the sonicator in isolating the optimum value of GSH was 15 sec and 84 μm (40%), respectively, where 18.13 μmol/ml GSH was isolated.
