Study of resistance in some advanced barley lines to Ustilago hordei in Khuzestan province with three inoculation methods

Many of cultivars especially hull-less barley genotypes are very susceptible to barley covered smut caused by Ustilago hordei (Pres.) Lagerh. On the other hand artificial inoculation of disease in order to genotypes screening is very difficult and also each used method for this purpose have some defects. So this study was conducted to investigation of some advanced lines and commercial cultivars with three different methods simultaneously in three years. In order to prepare of infected seeds in field, 27 genotypes of hulled and hull-less of barley were infected after pollination by spore spraying method in first year. Then this experiment was done in complete randomized block design in next two years. Three inoculation methods including only inoculation in field by spore spraying, inoculation in laboratory by spore suspension, and inoculation the infield, laboratory, and without inoculation as a check and 27 genotypes were considered as first and second factor, respectively. The number and percentage of infected spikle were recorded. Analysis of variance was conducted according to complete randomized block design and the means were compared by Duncan test. The results showed that inoculation method in field in complex to laboratory inoculation was effective method and also only laboratory inoculation caused significant infection in second stage. Lonely inoculation in field did not lead to appropriate amount of infection. In attention to easy inoculation, the most practical method is lonely inoculation in laboratory. In this study the MOLA/SHYRI//ARUPO*2/JET/3/… line was the most resistant and Jonoob cultivar was susceptible genotype.
