Diagnosis of disease bacterials of garlic and resistance evaluation of ten mass garlic to causal soft rot bacterial in Hamedan province.

In order to identification bacterial causal agents of garlic disease Hamadan province ( 2006-2008) samples of with symptoms soft rot of garlic cloves , yellowing (chlorosis) of leaves , dis colorations and numerous other abnormalities collected from different crops. From samples were collected , 56 bacterial straine purification and isolated . Samples were cultured on NA , Kings'B and EMB cultures. From the bacterials strains , 15 bacterial strains isolate and purificationed. Some of the strains on EMB media metallic green color colonies produced and some of the strains on Kings'B culture flourecent color colonies produced. All bacterial strains were gram negative , catalase positive , oxidase negative and glucose , fructose , galactose , cellobiose , melibiose , sucrose , ribose , manitole , manose , ramnose , arabinose used for growth and some strains were pectolytic and rotted potato tuber slices. According to the biochemical and phenotypic characteristics strains were identified as Pectobacterium carotovorum sub. sp. carotovorum and Pseudomonas syringae. In order to evaluate resistance of garlic plants to Pectobacterium carotovorum sub. sp. carotovorum inoculation with stem puncture method (sp). Garlic plants , Soolan , Tuserkan , Aliabad , Sheverin , Barfajin , Muein , Tuein , Bahar , Hydareh and Maryanaje each mass garlic three replicate were planted in green house. Thirty days after planting , the plantlets were inoculated with a suspension of the Pectobacterium carotovorum sub. sp. carotovorum ( at the ratio of 10 8 cfu per ml.) with the stem puncture method and check two plantlets were inoculated by distile water . Three weeks after inoculation , based on symptoms of wilting , yellowing , and necrotic of plantlets , by scale 0-5 (0 =non symptoms , 5= maximum wilting , yellowing and necrotic). Total analyses of susceptibility of mass garlic to P. carotovorum sub. sp. carotovorum under green house conditions such as mass Tuein , Maryanaje, Bahar, Aliabad , Soolan were susceptible and mass Muein , Hydareh and Tuserkan were relatively tolerance to P. carotovorum sub. sp. carotovorum . But mass Barfajin and Sheverin were resistant to P. carotovorum sub. sp. carotovorum.
