Establishment and maintenance of growth parameters for cell suspension cultures of Ficus deltoidea

Callus derived from the leafof Ficus deltoidea (`mas cotek`) female plant was used to establish the cell suspension culture in the shake flasks system. Different concentrations ofplant growth regulators were studied in order to produee rapidgrowing and well-dispersed suspension culture of F. deltoidea. Cell eulture was established in Murashige and Skoog (MS) media containing 3 mg/L picloram. Maintenance of suspension culture were examined in full strength MS media supplemented with 3% sucrose and the plant growth regulators; either 4-amino3,5,6-trichloro-2-pyridine (picloram), 2,4-dichlorophenoxy acetic acid (2,4-D)or dicamba at the concentrations of 1 to 5 mg/L. Packed cell volume (PCY) as well as cell number were measured at every three days interval. Results obtained revealed that MS medium supplemented with 3 mg/L picloram was shown to be the most efficient for establishment of cell suspension culture which gave the highest value of packed cell volume (PCY) and total cell count. The cell suspension culture of F delta idea was successfully established and maintained in this study. The growth curve constructed based on the cells dry weight showed that the culture was in their lag phase between day 0 to day 9. After day 9, the cells eventually entered the exponential phase and almost reaching stationary phase after day 21.
