Determination of diuron residue in soil by high performance liquid chromatography

In the present work, a method for the determination of diuron residue by HPLC-ultra violet in soil was developed. Extraction of the diuron residue from soil sample was carried out by liquid-liquid extraction with acetonitrile and followed by sonication. The optimum volume of acetonitrile for extraction of diuron was 40 mL and duration time for sonication was 20 min. Ten mL of acetonitrile extract was dried under nitrogen gas and top up with 1mLacetonitrile/water (1:1, v/v). HPLC separation was carried out on anAscentis™ RP-Amide column with elution of acetonitrile (solvent A) and water/methanol (2: 1, v/v) (solvent B) at ratio of 4:6 (v/v). A linear correlation of 0.9983 was obtained for diuron at concentration of 0.05-1.0 ugmL-l Limit of detection (LOD) and Limit of quantitaion (LOQ) values are 0.01 and 0.03 ugml·1. The recoveries of diuron from the soil with five spiking levels (0.2,0.4, 0.6, 0.8 and 1.0 ugg-l) were in the range of 93.2 - 98.7% with a relative standard deviation (RSD) in the range of 0.43 - 4.83%. Therefore, the method developed is sensitive enough for the determination of diuron in soil.
