Clarification of fermenter inactivated FMD virus with sartoclear filter for vaccine preparation.

Foot and Mouth Disease Vaccine is a killed vaccine that it s quality depend on the antigen clarification and concentration per dose and adjuvant. The FMD virus in one serotype (O) was cultivated in Baby Hamster Kidney cell cultures. Study on clarification (active FMD virus ) with use of Sartoclear filter with different Pore size (8um , 4um , 1um )and measuring other factors like TCID50% - Complement fixation unit antigen detection by ELISA and PCR in order to evaluate that is it possible to clarification the FMD viral suspension with filter instead of Continuous Centrifuge . That is the main subject of this project . There are two main method for Virus clarification, continuous centrifugation and filtration. After filtration with different Pore size as mention above ,suspension turbidity and viral antigen was assayed by quantitative complement fixation test- titration (TCID50%) ELISA PCR and 140s quantity. Complement fixation test, titration (TCID50%) , 140s quantity and virus also adsorption by aluminum hydroxide gel were used for evaluate of clarification . The results shows that 4um pore size filter is the best choice for clarification of viral suspension and can be substituted with continuous centrifuge.
