Map kinases SIPK and WIPK of Nicotiana benthamiana are required for alteration os systemic immune reactions during infection by Pectobacterium carotovorum

إنجليزي. Efficient resistance of plants to pathogenesis supposes development of system stability, preventing further infection extension. Pectobacterium carotovorum represents wide-spread plant pathogen, affecting different plant varieties. The main goal of the research appears evaluation of the influence of gene silencing of MAP-kinases SIPK and WIPK of Nicotiana benthamiana plants on activation of system stability within affection with Pectobacterium carotovorum. The article presents the results of measurement of systematic changes in expression levels of pathogenesis related genes in Nicotiana benthamiana plants infected by Pectobacterium carotovorum. Comparable level of gene expression in system leaves of Nicotiana benthamiana infected with TRV2::GFP, TRV2::SIPK and TRV2::WIPK 24 hour after infiltration with Pca cells’ suspension has been measured. The comparison with control group, infected with 10 mM of MgSO4 has been made. Contrary to the situation in infected leaves, there has not been induction of PR genes in systematic tissues. Expression of four PR genes has been significantly decreased. Virus induced silencing of the SIPK and WIPK genes in Nicotiana benthamiana have shown that these genes are required for the observed suppression effect.

В отличие от полученных ранее данных для непосредственно инфицированных листьев мы не обнаружили индукции экспрессии PR-генов в системных тканях. Наоборот, экспрессия четырех PR-генов оказалась существенно сниженной. Вирус-индуцированный сайленсинг генов SIPK и WIPK у растений N. benthamiana показал, что эти гены необходимы для системной супрессии PR-генов в ходе бактериальной инфекции.
