Screening of the global gene pool of spring wheat according to resistance to brown rust and identification of Lr-genes in some varieties and breeding lines | Скрининг мирового генофонда яровой пшеницы по устойчивости к бурой ржавчине и идентификация lr-генов у некоторых сортов и селекционных линий

There have been presented the results of the analysis of pedigree and phytopathologic assessment of spring wheat collection material on the basis of which the most promising varieties and lines possessing genes controlling the resistance to leaf rust were identified. The investigation was carried out in St. Petersburg in 2013-2014. At the 1-st stage the phytosanitary situation in the wheat fields was observed, the development of diseases was recorded. At the 2-nd stage the samples naturally identified by resistance were estimated in the hothouse conditions by artificially infecting the plants with a complex population of leaf rust consisting of pathotypes collected from the wheat crops in different places of the Saratov region. At the 3-d stage leaf rust resistance genes were identified by using molecular markers. There are received data, which allow to regulate the use of donors with identical resistance genes and to extend the existence time of resistant varieties. With the use of DNA markers for a number of Lr-genes identification of 8 Lr-genes was carried out (Lr3, Lr9, Lr10, Lr19, Lr20, Lr23, Lr26, Lr35) in varieties Danphe, Clez Alta and MN91095, as well as in introgressive lines of spring wheat (Saratovskaya 55 // Tr. dicoccum/Ae. speltoides, Saratovskaya 55 // Tr. dicoccum and Saratovskaya 55 // Lr9), which carry genetic material from wild species and relatives. It is found out that these lines have both effective and ineffective resistance genes. In varieties Picaflor, PRL/2 and the Morris, resistant to the Volga region leaf rust effective genes were not found, possibly due to the lack of molecular markers for the corresponding resistance genes. It is shown that the use of DNA markers for Lr-genes identifying allows detecting resistance genes that cannot be determined by other methods.

الروسية. Представлены результаты анализа родословной и фитопатологической оценки коллекционного материала яровой мягкой пшеницы, на основе которых выявлены наиболее перспективные сорта и линии, обладающие генами, контролирующими устойчивость к бурой ржавчине. Исследования проведены в Санкт-Петербурге в 2013-2014 гг. На 1-м этапе исследований проводили наблюдения за фитосанитарной обстановкой на полях яровой пшеницы, учеты развития заболеваний. На 2-м этапе образцы, выделенные по устойчивости на естественном фоне, оценивали в тепличных условиях при искусственном заражении растений сложной популяцией бурой ржавчины, состоящей из патотипов, собранных с посевов пшеницы в различных местах Саратовской обл. На 3-м этапе идентифицировали гены устойчивости к бурой ржавчине с использованием молекулярных маркеров. Получены данные, которые позволяют регламентировать использование доноров с идентичными генами устойчивости и в результате продлить срок существования устойчивых сортов. С использованием ДНК-маркеров для ряда Lr-генов проведена идентификация 8 Lr-генов (Lr3, Lr9, Lr10, Lr19, Lr20, Lr23, Lr26, Lr35) у сортов Danphe, Clez Alta и MN91095, а также у интрогрессивных линий яровой мягкой пшеницы (Cаратовская 55 // Tr. dicoccum / Ae. speltoides, Саратовская 55 // Tr. dicoccum и Саратовская 55 // Lr9), которые несут генетический материал от диких видов и сородичей. Установлено, что данные линии обладают как эффективными генами устойчивости, так и неэффективными. У сортов Picaflor, PRL/2 и Morris, устойчивых к поволжской популяции бурой ржавчины, не выявлено эффективных генов, возможно, из-за отсутствия молекулярных маркеров для соответствующих генов устойчивости. Показано, что использование ДНК-маркеров для идентификации Lr-генов позволяет выявлять гены устойчивости, которые невозможно определить иными методами.
