An efficient protocol for plant regeneration in eucalyptus urophylla viasomatic embryogenesis induction

A system for callus induction from hypocotyls and regeneration of Eucalyptus urophylla via somatic embryogenesis has been studied. Results showed that the rate of somatic embryogenesis formation on medium (MS reduced 1/2N supplemented with 1.0 mg/1 BAP, 0.1 mg/l kinetin, 0.5 mg/l NAA, 0.1 mg/l IBA, 20 g/l sucrose, and 2.0 g/1 phytagel) was 62,2%. About 70% of somatic embryos were germinated on MS reduced 1/2N medium supplemented with 0.5 mg/l BAP, 0.3 mg/1 NAA, 0.1 mg/l IBA 20 g/1 sucrose + 2.0 g/l phytagel under light source at lux values 1.500 - 2000 lux. The highest shoot multiplication factor (5.03 times) was obtained in MS supplemented with 0.3 mg/l BAP, 0.05 mg/l NAA, 0.1 mg/1 IBA, 20 g/l sucrose, 6 g/l agar, 0.3 mg/l vitamin B2, and 0.075 mg/l folic acid. Shoots about 2-3 cm in length rooted healthily on 1/2MS reduced 1/2N additional 0.3 mg/l NAA, 0.2 mg/l IBA, 20 g/l sucrose, and 2.0 g/l phytagel, Plantlets were transferred in soil pots with 95% survival rate.
