Molecular diversity of Grapevine leafroll-associated virus-2 (GLRaV-2) in autocthonous grapevine varities in Eastern Mediterranean and Southeast Anatolia regions

In this research, prevelance and molecular diversity of Grapevine leafroll-associated virus-2 (GLRaV-2) was investigated in autochthonous grapevine varieties in major viticultural areas of Eastern Mediterranean and South-east Anatolia regions. Research materials were green tissues stored at -20 oC and greenhouse-forced rooted cuttings from previous research. Leaf and petiole were used for total nucleic acid (TNA) isolation with silica capture method in GLRaV-2 detection. Two step polymerase chain reaction (PCR) was performed with virus-specific primers (LR-2F/LR-2R). Four out of 206 tested grapevine samples resulted positive in PCR and gave a DNA band of 543 bp in size. PCR DNAs were sequenced in two directions, aligned with CLUSTALW and analysed in Bastn for multiple comparison of other isolates in gene bank. Nucleotide sequence of the Turkish LR-2 isolates from grapevine showed 99% of identity with those from other LR-2 isolates. The Turkish LR-2 isolates were closely related to other isolates from Poland (JN865253.1), Italy (DQ314603.1) and South Africa (AY881628.1) in the phylogenetic tree prepared with neighbour-joining method in MEGA 6.
