Preliminary study on growth performance of Micheua alba callus cultures

A preliminary study on growth performance ofMichelia alba or known as 'cempaka putih' callus cultures was studied. Callus of M.alba was obtained by inoculating the M.alba explant (green petal of stage 5) on a basal Murashige and Skoog (MS) media supplemented with phytohormones, antibrowning agent and different concentration of casein hydrolysate (CH) (25, 50, 100 and 150 mg/L, w/v). The growth performance of of M.alba callus culture on two type of solidifying agents, namely Gelrite and Kelcogel was then studied. The growth profile of the callus cultures was monitored for 30 days with 6 days interval between each observation. In each observation, weight of callus culture was measured on dry weight basis. The growth of M.alba callus cultures in Gelrite was found highest at 30 days of observation in all the cultures that supplemented with 25, 50, 100 and 150 mg/L (w/v) casein hydrolysate. However, the growth of callus cultures in Kelcogel was only highest in 50 and 150 mg/L (w/v) casein hydrolysate at 12 days of observation with 0.106±0.02g and 0.097± O.017g, (dwt/culture), respectively. Therefore, in this study Gelrite is more preferable than Kelcogel prior to its high capability in propagating the callus cultures of M. alba based on dry weight basis per culture.
