Isolation, production and characterization of protease from Bacillus sp isolated from soil sample collected from the Northern Iraqi region

The material of this research; He collected soil samples from the northern Iraqi region. Bacillus isolates K1 and K2 isolated from a total of 10 soil samples were found to be positive for protease enzymes.Casein Agar was used to determine the ability of the isolates to produce protease. Petri dishes were left to incubate for 48 hours at 30°C and then examined on a dark background. The presence of a transparent region around the intensely growing bacterial colonies was assessed as positive for casein hydrolysis.Bacillus K1 and Bacillus K2 isolates were identified according to their morphological, physiological and biochemical characteristics and 16S rRNA sequencing analysis.In the phylogenetic analysis of 16S rRNA gene regions of five isolates of Bacillus genus; it was identified that in the compared region of 879 nucleotide., K1 isolate showed similarity to Bacillus cereus KC571175 with 5 nucleotide difference and 99,4 % similarity and both to Bacillus thuringiensis NR112780 and to Bacillus toyonensis NR121761 with 1 nucleotide difference and 99,8 % similarity. K2 isolate was similar to Bacillus cereus KC571175 with 5 nucleotide difference and 99,4 % similarity; to Bacillus thuringiensis NR112780 with 7 nucleotide difference and 99,2 % similarity; to Bacillus toyonensis NR121761 with 6 nucleotide difference and 99,3 % similarity.
