Increase in the detection sensitivity of ´candidatus phytoplasma spp.´ 16srx group by real-time pcr using rna as a source material

´Candidatus Phytoplasma spp.´ of 16SrX group cause serious diseases of fruit trees. Present work describes a novel method for the 16SrX phytoplasma detection that utilizes RNA instead of DNA as a source material in real-time PCR settings. The analysis was carried out in 20 fruit tree samples that had been previously tested positive either for ´Candidatus Phytoplasma mali´, ´Candidatus Phytoplasma pyri´ or ´Candidatus Phytoplasma prunorum´. Our results showed that the use of RNA as a source material could lead to an average 3.3fold and 4.2fold increase in the detection sensitivity for ´Ca. P. mali´ and ´Ca. P. pyri´, respectively, and to more than 18fold increase in case of ´Ca. P. prunorum´. A higher detection sensitivity then translates into a higher possibility of a pathogen detection in a plant material even when it is present in a very low concentration, lowering thus the probability of false negative results.
