The aim of the research is to identify the Pst races found in Latvia, to find out the cultivated wheat varieties response to aggressive Pst races, as well as the identification of Puccinia species in barberry and the role of barberry as an intermediary in the development of Pst in Latvia. Identification of Pst races by the phenotyping method on differential varieties and durability of wheat varieties tested in a greenhouse: air temperature 17–20 ˚C during the day (16 h), 9–12 ˚C at night (8 h), relative humidity 70–80%. An internationally recognized (Chen, Kang, 2017) set of differential varieties was used, which consists of 20 winter and summer wheat varieties containing different resistance genes In May 2020, 54 leaves and berries were collected on barberry throughout Latvia for the identification of Puccinia examples. Samples from wheat were analysed in parallel. Fungal DNA was isolated using the Qiagen DNeasy Plant Mini kit set. PCR amplification and sequencing of the ITS1 / 5.8S / ITS2 / 28S region was performed using primers ITS5, Rust1, ITS4, Rust3 (White et al., 1990; Driessen et al., 2004). Sequence analysis was performed with Lasergene 14. Obtained in full length, compared to the sequences available in the databases for the species identification. Phylogenetic analyses were performed with the computer program PAUP. Aggressive Pst races have been identified in Latvia: Warrior– and Warrior. Pst races Warrior and Warrior– are virulence on winter wheat varieties ‘Brencis’, ‘Ceylon’, ‘Fredis’, ‘Edvins’, ‘Magnifik’, ‘Olivin’, ‘Reinis’, ‘Skagen’, ‘Talsis’ and ‘Zeppelin’ are in the juvenile stage, while Triticale 2015 is avirulent. In Latvia barberry is mainly the causative agent of wheat stem rust P. graminis f. sp. tritici intermediate owners.
