Luisoni, E. | Roggero, P. (Consiglio Nazionale delle Ricerche, Turin (Italy). Istituto di Fitovirologia Applicata)
After some considerations on development of serological diagnosis, we describe the most common methods used at present for the detection of plum pox virus ("Sharka"): immunoelectron microscopy (IEM) and enzyme-linked immunosorbent assay (ELISA). We show the essential similarities between the two methods, despite differences in technique, and the modifications sometimes used. We also describe the precautions necessary for obtaining good results and the advantages and disadvantages of each method, and offer suggestions for choosing the most suitable method to be used in different circumstances
