Comparison of nucleic acid extraction and yeast extract preparation from baker's yeast Saccharomyces cerevisiae and fodder yeast Candida utilis

Six modifications of the extraction procedure for the yeast extract preparation and nucleic acid isolation from Saccharomyces cerevisiae and Candida utilis cells have been compared. For the yeast extract production the best procedure was the autolysis of Candida utilis at the temperature of 50 to 52 degrees centigrade without any chemical agent additions. The best extraction procedure for the isolation of nucleic acids has also been achieved with Candida utilis yeasts under the following conditions: in 0,4 mol/l NH4OH, at the temperature of 40 or 92 degrees centigrade and pH 7,5. With S. cerevisiae an increased content of sterols was found in extracted cells with all extraction procedures.
