Studies on the genotoxicity of Ochratoxin A in cell cultures derived from pig and sheep

The genotoxic potential of ochratoxin A (OTA) has been investigated in two model systems, i.e. porcine urinary bladder epithelial cell (PUBEC) and ovine seminal vesicle (OSV) cell cultures using a sister chromatid exchange (SCE) assay and an in vitro micronucleus assay, respectively. For OTA, a concentration dependent increase in SCEs was measured in (serum free) PUBEC cultures treated with the mycotoxin (100 pM-100 nM). OT-alpha, its supposedly non-toxic metabolite, increased also the SCE-frequency, but at higher concentrations. In OSV cells (routinely cultured with fetal calf serum in the medium) the cytotoxicity of OTA, a compound with high affinity for serum proteins, was affected by the FCS-medium content. A concentration-dependent induction of micronuclei was observed for OTA in cytokinesis blocked OSV cells. Analysis of OTA-induced micronuclei by means of CREST-antibodies revealed a mainly clastogenic mode of action. Together these results support the idea that OTA which has been classified by the IARC as a possible human carcinogen is genotoxic in mammalian cells.
