Optimization of multiplication phase in magnolia x soulangiana in vitro

Two-year trials have been established aimed at action of concentration of benzyladenine (BA): 0.0; 0.5; 1.0; 2.0; 4.0 and 6.0 mg/l on multiplication culture Magnolia x soulangiana. Furthermore, the effect of addition of medium by low concentrations (0.0; 0.2; 0.5 mg/l) of beta-indolylbutyric acid (IBA) in interaction with the type of illumination was studied. The following traits were investigated: the number of offshoots per explantate, length of offshoots (mm), performance (average number of offshoots x average length), leaf area index (leaf length x leaf width), colour of leaves and production of basal callus (diameter in mm). The best results were obtained with medium Mil. + 1.0 mg/l BA (average number of offshoots 4.6; average length of offshoots 5.0 mm). Performance of the culture was raised by addition of 0.2 mg/l of BA in all variants of illumination and best results were obtained at constant white fluorescent illumination (40 micromol/m** (2)/s). Higher dose of BA had a negative impact.
