Evaluation of wheat (Triticum aestivum) cultivars with standard Gli/Glu alleles using PCR and leaf tissue as a direct template

Preliminary studies were performed to prove, whether sequence-based primers could be used for gliadin/glutenin loci identification instead of polyacrylamide or starch gel electrophoresis. The possibility of evaluation of DNA-based markers using direct template was studied showing that a segment of tissue is usable for PCR reaction also in wheat tissue. Segment of leaf tissue is suitable, unexpensive template, wheareas coleoptile did not work for this purposes. Two sets of primers derived from gama-gliadin pseudogene were used to analyze a set of wheat cultivars with standard gliadin alleles (Gli-B1 locus). Using F1 x R and F2 x R primer combinations, 13 different bands were recorded, which made differentiation of 12 groups possible. Such reaction could be therefore a simple and alternative method for their verification.
