Superovulation and recovery of zygotes suitable for double-microinjection in three rabbit populations

This study investigated the effect of superovulation in various populations of rabbits and different time interval between mating and egg recovery on the recovery of zygotes suitable for double-microinjection. Superovulations were used successfully to enhance the number of embryos and double pronuclei injection is more efficient to produce a large number of transgenic animals than single microinjection. Therefore, using these method we can reduce the cost of transgenic rabbits. We found, that in New Zealand White, California and Zobor population eggs recovery per donor resulted in 26, 32 and 21.8 zygotes which were suitable for double-microinjection; the time interval of 19-20 h between mating and recovery to obtain suitable zygotes was found to be a sufficient and the proportion of zygotes selected for morphological criteria (present of the pronuclei, polar bodies, intact zona pellucida and compacted cytoplasm) did not vary (F-test) among the rabbit populations (87.05, 88.72, 88.23).
