Cereals, especially wheat, had been considered recalcitrant for a long time. Different plant tissues were tested for their regeneration ability in vitro like leaf segments, roots, floral segments, mature and immature embryos, pollen and microspores. Agrobacterium does not attack monocots in nature, which is the second reason why transformation of cereals is more difficult as compared with dicots. There were effort in some laboratories to manipulate Agrobacterium genome to force it to interact with monocots. Some phenolic compounds like acetosyringone were added to the cocultivation media to induce Agrobacterium virulence regions. Other types of transformations were tested as well. Particle bombardement were proved to be the most successfull for all the cereals.
