Analysis of asymmetric intergeneric somatic hybrid plants produced between Hyoscyamus muticus L. and double mutant of Nicotiana tabacum L.

إنجليزي. Intergeneric somatic hybrid plants were produced between Hyoscyamus muticus and a double mutant of Nicotiana tabacum (nitrate reductase deficient (NR-) and streptomycin resistant (SR+)). The hybrid showed chromosome instability and resembled in plant morphology to H. muticus. The somatic hybrid clones showed presence of peroxidase bands from both the parents. Fraction-1-protein analysis showed the large sub-unit of the somatic hybrid corresponding to N. tabacum, indicating that the cytoplasm of the hybrid plants were essentially that of tobacco. Such nuclear-cytoplasmic combination resulted in altered flowering behaviour, induced in vitro flowering and showed changes in flower morphology. The hybrid plants were completely sterile under selfed conditions but upon back-crossing with H. muticus some seeds were obtained from which mature plants could be obtained

[Sono state prodotte piante ibride somatiche intergeneriche fra Hyosciamus muticus e un mutante doppio di Nicotiana tabacum (carente di nitrato riduttasi [NR-] e resistente alla streptomicina [SR+]). L'ibrido presentava instabilita' cromosomica e assomigliava, per la morfologia della pianta, a H. muticus. I cloni dell'ibrido somatico evidenziavano la presenza di bande di perossidasi derivanti da ambedue i parentali. L'analisi delle proteine della Frazione-1 mostrava la grande sub-unita' corrispondente a N. tabacum, indicando che il citoplasma delle piante ibride era essenzialmente quello del tabacco. Tale combinazione nucleare-citoplasmica si traduceva in un comportamento alterato alla fioritura, induceva la fioritura in vitro con un cambiamento nella morfologia fiorale. Le piante ibride risultavano completamente sterili se autofecondate, ma, a seguito di incrocio di ritorno con H. muticus, sono stati ottenuti alcuni semi da cui si sono potute ottenere piante mature]
