Development of a rapid procedure for in situ evaluation of Saffron colouring strength, comparison with HPLC and spectrometric procedure

T Hunter color parameters (L*, a*, b] were measured with a portable tristimulus colorimeter and chroma and hue angle were calculated with high precision (CV per cent 0.01-0.11 for all parameters). Saffron aqueous extracts were used for spectrometric measurement and HPLC characterization of water soluble carotenoids, responsible for the coloring strength of the spice. Univariate regression analysis showed a good correlation between absorbence readings expressed as E1 per cent per 1cm and red chromaticity, a* (r2=0.782). Multivariate regression analysis improved the predictability of the linear model for absorbance readings. A satisfactory correlation between colorimetric and spectrometric data was found (r2=0.831). The same crocin pattern was recorded for all samples when RP-hPLC was applied to saffron aqueous extracts.
