RAPD analysis of two pairs of su1se/su1Se near-isogenic lines for the identification of chromosomal regions affecting the sugary enhancer phenotype [Zea mays L. - sweet corn - random amplified polymorphic DNA]

Sweet corn with the sugary enhancer (se) phenotype is traditionally characterized by elevated sugar content, slow dry down of kernels, more wrinkled pericarp of mature dried seeds and lighter kernel color. The objective of our study was to identify chromosomal regions which affect these characteristics using RAPD analysis of near isogenic lines (NILs). Two independent pairs of near isogenic lines (IL678a and IL451b) were compared. The original sulse line IL677a was used as a reference. Three hundred and forty arbitrary, 10-mer primers were used to amplify genomic DNA extracted from the different genotypes. Three of the 340 primers generated amplification products which differed between the sulse genotypes and their sulSe isolines. The association between the RAPD discrete bands and the sugary enhancer phenotype was confirmed by analyzing 16 additional sulse and sulSe sweet corn inbreds. We mapped two of these markers to chromosome 6 and one of them to chromosome 3 by the analysis of their segregation in an se x Se F sub 2:3 population. The marked chromosome 3 region had been previously reported as being associated with kernel sugar content and taste, while the chromosome 6 region was associated with both maltose concentration and kernel color. The results indicate that more than one locus may affect the sugary enhancer phenotype. The complications in phenotypic selection and the advantage of marker assisted breeding (MAB) for the se phenotype are demonstrated

[Il mais dolce con il fenotipo miglioratore della zuccherosita' (se) e' tradizionalmente caratterizzato da elevato contenuto di zuccheri, essiccazione lenta delle cariossidi, pericarpo piu' rugoso dei semi maturi secchi e colore piu' chiaro della cariosside. L'obiettivo del nostro studio consisteva nell'identificare le regioni cromosomiche che influenzano queste caratteristiche utilizzando l'analisi RAPD di linee pressoche' isogeniche (NIL). Sono state confrontate due paia indipendenti di linee pressoche' isogeniche (IL678a e IL451b). La linea originale su1se IL677 e' stata utilizzata come riferimento. Per amplificare il DNA genomico estratto dai diversi genotipi sono stati impiegati 340 primer decameri arbitrari. Tre dei 340 primer hanno generato prodotti di amplificazione che differivano fra i genotipi su1se e le loro isolinee su1Se. L'associazione fra le bande discrete e il fenotipo miglioratore della zuccherosita' e' stata confermata mediante l'analisi di altre 16 linee consanguinee su1se e su1Se di mais dolce. Abbiamo mappato due di questi marcatori sul cromosoma 6 e uno sul cromosoma 3 mediante l'analisi della loro segregazione in una popolazione F inf 2:3 se x Se. La regione del cromosoma 3 marcata era stata indicata in precedenza come associata al contenuto di zucchero e al sapore della cariosside, mentre la regione del cromosoma 6 veniva associata sia alla concentrazione di maltosio, sia al colore della cariosside. I risultati indicano che piu' di un locus puo' influenzare il fenotipo miglioratore della zuccherosita'.
